Hi Randy, welcome to our site.
We’ve have a number of discussions around AKG in the past - I recommend you check them out:
People have discussed sodium butyrate here also, but they are scattered all over the forums, as you can see from our search feature (top right of the page): Search results for 'sodium butyrate' - Rapamycin Longevity News
We haven’t discussed Forskolin much here, what research has made you interested in it?
One short discussion here: Using rapamycin in bed bound elderly - #25 by Paul_2.0

I was intrigued to the combination by the the not very popular here David Sinclair’s anti aging cocktail and have been using weekly with lithium metformin and telmisartan I feel the boosting effect from these is worth
Experimenting with.

What are the ingredients of the cocktail? I’ve seen so many I’m not sure what you’re using.
I’ve taken AKG and Sodium Butyrate. The former for a couple of years. I took Rejuvant, with no discernible feeling of anything good or bad. I occasionally take Sodium Butyrate, which is a HDAC inhibitor, but I’ve taken it primarily to increase butyrate in my gut. I’m not tempted to take any of the combinations from Sinclaire’s lab. Too early for me.

There is some synergy at play when i combine
Forskolin sodium butyrate akg and tranylcypramine with Rapa. I feel more higher energy
Level and a feeling of euphoria.
Will go for blood work soon to be sure.

It’s interesting… We get quite a few reports of euphoria. See this thread and the links Euphoria after rapamycin
What would be interesting is if you experience the euphoria if you only take the rapamycin and not the other compounds.

Do you have a link to the David Sinclair cocktail? Would like to see the mg content per ingredient.
Thanks in advance.

No dosing details here though
Thank you.
Looks like we discussed this paper earlier.

As far as I know, chemical partial reprogramming has not been tested in whole mammals. Most of the work (such as the paper you link) is in vitro, although at least one paper extends to nematodes. Extreme caution is recommended here. Most uncommitted experimenters will be deterred by the lack of cheaply available TGF-beta signalling inhibitors.

Yes I think lithium and metformin do affect tgf beta to a smaller extent. I am assuming Most of these cocktails are targeting microbiome since each of us carries a different version of it, there might not be one size fits all. Mostly trial and error but biomarkers should lead us somewhere.

Did you ever tried taking AKG and butyrate with rapa if so What was your experience with that.

Yes. I take all three. Although, I don’t take butyrate all that often. I usually take it a few times a week if I remember. I haven’t really noticed much of anything. I think without tracking it’s hard to notice any kind of difference

Have you experienced any sort of synergistic effect by taking all three together
