You should have titled this thread “apprehension about taking an experimental drug”.
Yes, most people have it, for me reading the research trials on rapamycin is as soothing as a pacifier to a baby.

That’s such a great title - I think it developed into that for sure. I wonder if we can rename it because it makes much more sense. I’m a writer so I really appreciate the right title :slight_smile:

Fwiw, my cgm snapshot at any time during the day after I’ve eaten does not line up with my fasted blood work. So I’m not sure this says your glucose has risen as a result… but can’t say for sure!

After a long illness acquired from granddaughter’s daycare (she doesn’t go anymore) I had to take dose #1 again and all the apprehension is just a little less than the actual #1 :grin::grimacing:

I took the 4th dose yesterday (4 mg). The usual “apprehension about taking an experimental drug” kicked in this morning so I am here soothing myself being among fellow takers of said drug. I have a lot of joint pain in the joints that are poor to start with, but I’m hoping it was from my 9th lift number 1 (trying to get motivated to become consistent the way I used to be a year ago). Also I woke up with a allover tension/ neck type headache the last two times after the dose. I’m not saying this is caused by the dose, but? Also nausea, very mild but noticeable, started (hours) after the 3 mg dose. Tired all the time, sleeping a LOT - 9-10 hours.

Hoping all this disappears, i guess hoping to turn into @Agetron is asking far too much :grin:

I also took my weekly rapamycin/zydus dose last night… 4 mg with one fresh squeezed red grapefruit juice - is about 5 fluid ounces of GFJ.

This combo dose gives me an equivalent of 19 ng/mL or approximately an 8mg rapamycin/zydus dose without GFJ.

Within hours, I usually get a strong ringing in my ears - tinnitus. I usually take my dose at bedtime and can sleep through the night, but actually wake fairly groggy. A little foggy minded and unsure footing and unmovements.

You have to consider you just bombarded 38 trillion cells with information they’re all starving… Every cell type in every part of your body. You should expect to have a shock reaction.

Which wears off by about noon for me… which is 12 hours post dose. Now it is lunch time and I’m back to normal. The tinnitus is lesser over the next days… until my next dose.

No other negative effects. Actually starting to feel very euphoric and energetic. Maybe an alert to the whole body to get going and survive. I avoid gym and anything strenuous for a day and a half post dose.

Not sure what to say about the other aspects you are encountering. Perhaps when you next dose, you can see if they repeat or was it something else.

On the positive side, I can honestly report that after 3 years and 8 months of use…my skin quality, my oral health… my strength, my memory in all aspects… free of allergies…improved sinuses… generally free from illness. It’s as if I went back fifteen years in age. Fantastic.

Thanks for your posts, because you talk about the positives, which are many for you, but also these weird things like tinnitus (I have had a clicky ear since my 3rd dose, which was the one that had any real effect that I could not attribute to placebo/nocebo). I should keep a log, that way patterns will be evident when the weeks and months pile up. I hope to be able to report “Fantastic” after 3 years and 8 months :slight_smile:

The first time I took a “normal” weekly dose of 6mg I felt almost euphoric for the first couple of days. The next time I took a weekly dose I felt slightly tweaked as if I had drank too much coffee. Then the third time I felt sick and tired for 3 days (even though I lowered the dose to 5mg every two weeks). Think I will be taking a more extended break now, maybe up to a few months.

This is the bottom line. Long term use leading to improvement in the conditions of aging, the short term effects either somatic or real are are unlikely to be clinically relevant, except for occasional infection or other rare but more serious side effects.

I’m getting ready to post a 4 year review. All biological tests (inflammation…glycan; methylation), new coronary calcium scan, dexa, current blood panel and photographs that detail of all the things that have improved and correspond to my tests.

Will encourage others who have similar material to do the same… a rapamycin in review. My N=1.

I’m actually excited to see these! Really. I’m sure we’re all hoping that all your results are superlative, of course for you, but if we’re honest, it’s an inspiration and relief and validation too :smile: I took the 5th dose today. I read your posts to first calm down and then feel great about taking it.
