Reading through the interesting feedback above, last Friday, I had my first 5 mg take of rapamycin (Rapamune) and noticed a big increase in blood pressure. I have been taking 5mg of Bisoprol (beta blocker) in the evening and half a pill of Valsartan (angiotensin II) in the morning for couple of years now and my blood pressure was always in the 120/80 range. No later than 24 hours after my first dose, it went to over 156/95 and has been varying ever since. This was accompanied by a persistent headache for half a day.

@shtono Last Friday I also had a big jump in BP and headache most of the day. I took only 0.5 mg of Rapa. However, I donāt blame it on Rapa, but on atmospheric pressure that was abnormally high. BP could be seasonal - better in summer and worse when cold comes. At least itās true for me.

Thereās something there. It doesnāt seem to affect most people, but for some situations there seems to be something.

My BP systolic increased 10 pts this week. The previous week it increased 6 pts. The previous week was a decrease of 3 pts.
No solid pattern but it appears to increase 5-10 and then fall back to normal over 3 days.

@LaraPo This is very interesting and in fact, a healthy friend of mine complained of an increase of blood pressure, which he relates to atmospheric abnormalities during that period. My plan is to stabilize my BP and then start again with a low dose of 1 - 2 mg per and closely monitor BP.

More support for this wrist one - that has five clinical trials also from Peter Attia.
@John_Hemming it does measure during sleep.
Did you ever look into this one? Did anyone try it?

I am trying it. I also measure using a normal Omron. I am not yet persuaded to see any major value in the measurements. However, I will stick by it. It is really good in how it works, but its results may be out by say 15% or more. I donāt know.
I like the design and I will continue to wear it as it is not invasive and does not cause me to wake at night.

Is the Aktiia specifically that you are trying?
Yes it is an Aktiia software version 1.71 which I put on a few weeks ago. Next calibration is November. It looks very well designed. I just think the task is a bit too hard. I retain an open mind to the extent I continue to charge it.
I have just measured my BP at 126/79-70. Aktiia has one measurement in the period 12 noon to 2pm at 145/97-67.
Now I am expecting my BP to be a bit high (I have just done another Omron at 130/79-70) because I am simultaneously increasing acetyl-CoA in the cytosol and aiming both to increase the efficiency of the electron transport chain and also using other techiques to increase ATP levels. This I expect to put up my heart rate and blood pressure (I am also sober and did not drink yesterday which would normally put my heart rate around the 55 mark without all the other things).
The objective to this is improvements to mRNA transcription and also improvements to translation of mRNA into proteins. Because I took 6mg of Rapamycin yesterday morning I want to maximise the efficiency of autophagy although I am not fasting today although I did yesterday.

Seems like a transient increase in blood pressure is a real thing with Rapa judging by the number of people with very similar stories in this thread. I wonder how many Rapa takers just havenāt checked? Is there a way to to generate a poll to determine how frequent this is? Rapa dose, Rapa dose interval, BP days 1-3 or something like that? Iām surprised it hasnāt been previously described but most published studies donāt involve high intermittent dosing.
We are doing a multiple n=1 data collection on this.

For what itās worth, here are my blood pressure readings. Iāve just started using an Aktiia 24-hr blood pressure bracelet but also use a Withings blood pressure cuff twice a day.
Iāve just started a new dosing regime, where Iām taking 1mg of rapamycin twice weekly, sublingually (every Monday and Friday). if I decide to go to higher doses I might go to once weekly dosing.
So in the graph shown above my first dose was on the 10th and second dose on 13thā¦.and Iāve seen no increase in blood pressure.

Really good BP. Do you use any BP lowering medication? (Except SGLT2i which has the potential to reduce BP slightly)

The only meds I take that may impact on blood pressure are 10mg Telmisartan and 2.5mg Nebivolol. I know they are low doses but it seems to work for me.
Iām thinking of simplifying my meds stack soon and may see what I can drop. Itās all too easy to keep adding new meds.

Have you been taking these the entire time of the blood pressure readings you posted above? I love the results from the Aktiia monitor!
My aktiiia results go all over the place and donāt correlate with Omron adequately. OTOH I think they have done a good job of a really hard task. Hence I intend continuing to wear the strap. I have not so far IMO got any useful information from it, but it is well designed with a good piece of software. Hence I want to give it time.
FYI my current Omron is 113/68-57 and Aktiia 140/92-58. I am, however, slightly drunk so my BP will be a bit lower.

Yes, Iāve been on those meds for a while now.
Iāve seen no reliable pattern so far. Usually BP is higher the next day. Sometimes itās lower. Sometimes my BP is higher on any random day for no known reason. For me, I donāt think rapamycin vs blood pressure is a thing.

Iām not sure if youāre US or Europe/UK, I live in the UK.
I was bidding for the Aktiia bracelet on eBay but it went over my limit, then luckily I saw one on Gumtree for Ā£100, only used for 24 hours. They last for days before they need charging and charge quickly.

I think they go for about Ā£200 but am not sure. I think they are not for sale in the US yet, but I think the company may be be submitting it to the FDA shortly for accreditation.
