I just started taking Rapamycin couple of days ago for Anti ageing purposes. I am took 3 mg on Monday. I will take 4 mg next Monday, and 6 Mg Monday after (my target dosage). The protocol I want to follow is 6 mg once week.
Should I continue this protocol (6 mg once a week) indefinitely, or do some of you cycle…that is go for periods without rapamycin (rapaholiday). Maybe every 4 to 6 months go on a 1 or 2 month of taking no rapamycin (even weekly). What are some of protocols for cycling that you follow and is there any evidence for it? I also take Metformin 1000 Mg daily. I started it for anti ageing but now it appears like I am prediabetic (5.7 H1C haemoglobin). Any other drugs that have good synergy and/or ameloriates some of the possible negative effects of Rapamycin (like Skin cancer risk, Sacropenia, kidney etc).
Also I am thinking of going on Seracycine for Acne. Will this react with Rapamycin any toxic interactions etc? I heard Matt Kaberlin recently had an allergic reaction.

Hi and welcome to the site,

Rapamycin cycling (or “rapamycin holidays” or breaks of weeks or months), are something we’ve discussed a lot here. It seemed to be more common a few years ago and I think a lower percent of people do it now, but either way there is not much research on the issue, at least in healthy humans. So its going to be an individual choice based on the limited data (mouse models), and your personal reaction to rapamycin, side effects, blood work results, etc.

You can see some of the past discussions on this here:

Regarding drugs that help minimize or mitigate any side effects… many people here take acarbose or metformin, others take canagliflozin or other SGLT2 inhibitors (for their own benefits, but also to prevent any blood glucose increases), and others take statins or other lipid reduction approaches (again, for their own benefits, or for any lipid disregulation that may occur with rapamycin). More details on these approaches here:
