Preparation and quality evaluation of rapamycin suppository
Objective: To prepare rapamycin suppository, and to evaluate its quality. Methods: Rapamycin was used as active ingredient, polyethylene glycol 2000 and polyethylene glycol 4000 were used as main substrates. The suppositories were made using hot melt process method, and the drug content, weight difference, melting time, drug release with time were evaluated. Results: Polyethylene glycol suppository with rapamycin appeared white and translucent. Good hardness could be maintained at room temperature. The drug content, weight variation and melting time limit matched the requirements for the suppository. More than 75% of the loaded drug in the suppositories was released at about 25 min in the medium. Conclusion: This preparation of rapamycin polyethylene glycol (PEG) suppository can reach the quality requirements of Chinese pharmacopoeia, and may be developed as a new formulation of rapamycin.

Thanks for posting what Mr. Google found! I wonder if it would work to not use PEG 2000 or PEG 4000 and just insert the cellulose capsule containing the rapamycin.

You might also use Transcutol to increase absorption… its great for that; I use it for rapamycin skin cream and toothpaste. You want something like PEG or Transcutol to improve absorption.
Transcutol has a number of different chemical names/identifiers. Here is a list:

I tried the Purecaps product and got the same result that I got with Acid Armor: a blood test result of < 0.5 ng/mL Sirolimus (rapamycin). However, at that time I was taking a turmeric product (Meriva, turmeric phytosome), which might have skewed the result. Elsewhere on this site, I believe RapAdmin posted a paper showing that turmeric in the system causes the liver to detoxify or break down everolimus, which is similar to rapamycin. But I would have taken the Meriva at least 24 hours prior to taking rapamycin in a Purecaps capsule. The half-life of Meriva in the blood is reported on some websites as 7.5 hours. So I would have taken the rapamycin about 3.2 half-lives after taking Meriva, so the Meriva concentrations should have been only 11% as much as the peak concentration.
David, welcome to the forums. Is this delivery method (rectum) something you have personal experience with, regarding rapamycin? Did you do any testing to see how well it worked?
I am aware from posts in this forum that siraboon does have good purity and presence of Rapamycin . However it seems from blood tests that oral doses do not give the desired results due to a sub standard enteric coating I can only assume . So given this , I think that PR may work . I don’t have access to rap blood tests , so is anyone able to do the honours with PR SIRABOON ? I have no special expertise or experience by the way apart from what was in the link I posted previously .

lol, i do not want to that way lol. I use a separate enteric capsule to protect the pills. Cost like $15 for 200. I can easily put up to 3 pills into one.

Fair enough . But from what I’ve read , bioavailability is possibly many fold more than even the enteric capsule route . So was just wondering if someone had done the blood work PR , so if people do want to use that route , they can optimise dose

What brand are you using? Have you done blood testing an hour or two after consumption, to verify the rapamycin made it past the stomach and into the small intestines? I’m considering these; quite a bit more expensive, but still cheap compared to the hard pills. ~$50 for 100 size 4 empty capsules, and they’ve been tested.

Okay… I heard of this the other day in Palm Springs, CA… don’t ask how…hahahah! And, yes - my response was WHAT???
Booty Bumping - slang for taking drugs in your rectum (typically cocaine).
But heck… why not rapamycin and do a Labcorp test to see how much was absorbed into the blood stream.

I have published my blood results somewhere in the forum … have done over 30+ blood test just for siromilus alone. I do not plan to use the enteric capsule anymore as the results shows that the timing of taking the pill affects the siromilus level in your blood more. I prefer to do the blood test at day 3 or 4 days to see that it is above the 3ng/mL threshold. One of my blood test after 12 hours was over 40ng/mL.
Last week I had 10mg with GFJ. My 85 hour test is 4.7ng/mL. So I can safely say that on day 4, it should still be higher than 3ng/mL, may be even into day 5. Anyway, that is enough and hopefully my health will keep getting better.

I have just taken the Rapacan Biocon pills, and when I broke them out of the blister I was surprised to see them scored in the middle, implying you could cut them in half. These are 1mg pills. Why would you encourage people to cut the pill in half - wouldn’t that obviate the enteric coating? On second thought, the pills don’t have a particularly shiny surface, so I wonder about the enteric coating in the first place. This is from Jagdish, and an established brand. Ultimately I want to test my blood levels of rapamycin, because there is really no other way to know.

I think it is the moulding they use, I won’t split it in two. Take longer breaks instead and see if your body gets use to it.
