Is ageless testing you for Sirolimus?

I think this is just generic blood test before they will give me the free drugs?

I got mine and it specifically says sirolimus is one of the tests. Did u let them know u we’re taking it from India?

After reading the 33% reduction in Viseral fat with the compounded it sounds encouraging regarding the compounded. I’m gonna try the generic first and they said I could switch later if I want.

Just checked sirolimous is on their bloodwork panel, I don’t think they asked if I was already on it…

I’m sure it’s no big deal.

I thought that was strange to have on a prelim blood panel too. In my case, I continued on with the generic sirolimus after the trial for a few months. Got up to 8mg dose, which made me feel “off” “weird”, rather difficult to describe. Didn’t like it so I quit. Going to start up again with the compounded to see how it goes. I felt nothing on the trial and correctly concluded it was a placebo.

Good to know about the generic. I will see how it goes for me. Did you feel okay before you got to 8mg? Mine is on its way to me. They’ll start me with 2 mg. Keep us posted with the compounded.

AgelessRx did not test sirolimus levels during the PEARL trial for a variety of different reasons.
However, a couple months ago we started checking sirolimus levels on everybody who gets prescribed rapamycin.

The challenge is that not very much is known about the correlation between Sirolomus levels and outcomes. So right now, we’re not doing anything with the information other than aggregating it to help us better understand how to dose rapamycin

Also, I spoke with Sajad in December when I saw him at the Longevity Summit at the Buck Institute. He’s not committing on a date for the data… they’re working with a number of different groups who have all the pieces of the data from the testing and they need to pull it all together and compile it and analyze it.

And - they are a small company doing their regular business, so the PEARL study is a non-profit project they are doing on the side.

So… maybe if we’re lucky the second half of the year?

Specifically, how were those administered? weekly? daily? divided? I did not find that specific info on their website.

Shame they didn’t look at fortnightly dosing but when you are trying to power a small study with limited funds I understand why they didn’t.

I’m cautious about citing other side effects or benefits, since they are a lot less objective than a before-and-after DXA scan (same DXA machine and technician each time) that showed the reduction in visceral fat. I have not had any canker sores or similar issues. During the 12 months of the PEARL trial, I didn’t “start or stop anything else” related to anti-aging, but now that the trial is over, I’m taking low-dose Metformin (just 250mg daily) in addition to once-weekly 10mg generic Rapamycin, and varying my supplements modestly.

I think you have most of this correct.
We decided to go with 2 dosing arms: 5mg weekly and 10mg weekly (and a placebo).
All arms were using compounded versions. As dicusssed on this site (maybe somewhere on this thread), there is a lower bioavailability for the compounded version (we are publishing a seperate paper soon on compounded vs generic)
