I was hoping that some of you with a far more scientific brain than mine can help me sort some queries.

I need to know if topical sirolimus could potentially affect the healing of a cosmetic procedure and cause problems. I am referring to invasive procedures such as medium depth chemical peels + ablative lasers (or even less invasive procedures). I had a medium depth chemical which resulted in some very mild scarring and redness. I recommenced usage of topical sirolimus at approximately 14 days post peel. This outcome may be completely unrelated but I’m curious. I’ve never had healing issues before and I’ve only been using this a couple of months.

Also, I am considering taking oral Sirolimus so will this potentially cause problems when recovering from these this of invasive procedures which I plan to continue doing?

There are really very few clinical studies on topical rapamycin, so a short answer is that we don’t know for sure. But there are studies on rapamycin (orally delivered, daily) and wound healing and yes, as the dose gets higher there is a delay in wound healing, so it is possible. To be cautious, it would seem to be a reasonable strategy to delay rapamycin use topically while the skin is healing.

and while the doses that are typically used in longevity are low and only doses once per week or once every two weeks (depending on your specific dosing protocol), compared to the daily use in rapamycin’s typical use case (organ transplant patients, or cancer), there is reason to believe that it could potentially impact wound healing and recovery time.

Most of us here take a cautious approach and pause rapamycin use during any surgical procedures or when we are healing from an injury.
