I am starting rapamycin today.

Like another user who posted recently, the past year and a half I have been taking GLP1 medication, which led me to other peptides, and this led me to rapamycin. I’ve lost more than 100 pounds, I have 18 more to go… And today I plan to start rapamycin.

Currently I also take tirzepatide and pentosan. I am planning a cycle of several bio regulators starting in January, including a cycle of Fox04-DRI.

My plan is to start rapamycin at 2 mg, increasing by 2 mg each week if there are no side effects. Is there anything else I should know or be doing as I start?

Thanks for this forum, your knowledge, and anything you can suggest.

Have you done some reasonable blood testing to get baseline levels of all the regular measures? Its always good to know pre-rapamycin levels, so you can compare them later to post-rapamycin levels.

Needs clarification.

Maybe you get a mouth sore. (Perhaps most common side effect.) Are you going to stop? No? Then escalate. Yes,? How much do they bother you? Maybe maintain dose for a couple of weeks, and see if you get more.

What is your LDL level? Another common experience is increased LDL. Do you care? Plan to start ldl-lowering meds if you care. Otherwise, carry on.

Starting multiple things in the same timeframe makes it difficult to determine the culprit if you experience untoward effects. In my experience.

I have read mixed reviews in the topic.

You’d decrease your risk for stroke and heart attack if you take cholesterol* lowering meds significantly. The longer period you’re on them the further the risk lowers. Diet changes help by replacing saturated fat with polyunsaturated and monounsaturated fats (like canola oil and olive oil, and eating less of saturated fat in general). Right now your risk is higher and you continue to develop atherosclerosis, at an accelerated rate.

*By lowering atherogenic apoB lipoproteins.

On a general note, you will witness when reading and researching this that many people go on about total mortality with regards to lowering serum cholesterol, but they are often disingenuous. Strokes and heart attacks are terrible even if they aren’t fatal. But statins lower total mortality, anyway, and apoB (LDL cholesterol) reduces chances of living a long life by a lot, especially when elevated: Apolipoprotein B (ApoB).

I started in April and i followed an approach similar to yours… first week I probably took 1 mg… then raised it to perhaps 3… then quickly got to 6.

Apob is a great thing to follow if you don’t already.

Many people here take statins
Many are also on ezetimibe and bempadoic acid
Some even on repatha
If I tolerated statins, my docs would happily have me on them
Lots of options.

FWIW, I’ve had heart disease forever and rapa has not affected my levels. I did do blood work pretty quickly after starting and repeated again after a little more time, just to be sure.

Also a huge congratulations on your weight loss. You must feel great and also proud!

Blood tests before and while taking rapa, Dexa scan, establishing your own health goals and tracking them. Most importantly having realistic expectations and not thinking of Rapa as some miracle drug that it will make you 25 again. it can’t and won’t do it.

Ah yes - I forgot that. This is actually really valuable as in the PEARL study they were using visceral fat as one of the endpoints so you should see it decrease over time, and it would be great to have another data point for all of us here.
