Eventually Ageless and Healthspan will have pretty good info on rapamycin and the biomarkers they routinely test. They could conceivably invite patients to share their EHRs to further enrich their data. It would just be observational, but could offer some insights.

Very good point.

We will need to lobby Ageless and Healthspan to get them to release this information (anonymized) ā€¦ perhaps partner with an academic group that would take the data, combine it, and publish something together to help move the industry forward faster.

@szalzala and @Daniel_Tawfik What do you think - any chance we could get your companies to share some anonymized patient data and lab results, to help identify the dose / benefit tradeoffs with rapamycin as used by your clients?

We could probably get money from the https://impetusgrants.org group to fund the evaluation, summary and publishing of the data. Iā€™m happy to co-ordinate this effort if you guys are up for it.

My dosing of everything I take including prescribed medications is a best guess either by my doctor or me,
I adjust my dosing based on my research and I try to keep current.
My stack is constantly being updated as per dose or efficacy.

I have been taking Rapamycin for almost 2 years now. At first, I had a euphoric fatigue around dosing time when I was taking about 9-10 mg eq. a week. After a while, on the same dosing, the fatigue disappeared and it became more of an ā€˜energizedā€™ feeling for a couple of days. It coincided with my addition of daily taurine, so that could be the key.

For several months now, I have moved on to a higher dose of 14 mg every 2 weeks and that euphoric fatigue and energized feeling are both gone and I generally feel fatigued for a few days after dosing. However, during the second week, my body feels energized like before. This is my N=1 experience and Iā€™m not sure what conclusions I should draw from this.

My biomarkers for HBA1C (5.4) and lipids (LDL 65) are all in (almost) optimal ranges after the addition of daily Bempedoic Acid, Ezetemibe and Metformin (500 mg).

Maybe I need to go back to 9-10 mg eq. a week or stay at a higher dose on a bi-weekly schedule?

Looking at HED doses extrapolated from the trials in mice , doses of 1-2 mg per kg , it is a DAILY dose of 6mg for a 75 kg person .

I think thatā€™s way too muchā€¦ at least for me, based on my biomarkers. 6-8 mg weekly works fine.

Thatā€™s what I take for general health and longevity , but Iā€™m extrapolating from the trials using BSA algorithms from specifically the trials for periodontal remission in mice where doses of 1g/kgper day *3/37 , multiplying this by 75 kilos for me I get 6mg per day (for periodontal remission)

Bi-weekly (fortnightly) makes more sense but I could be wrong. If we eat enough red meat, we do not need taurine right?

Please be clear when you use the term ā€œbi-weeklyā€ that you mean every two weeks (some people interpret ā€œbi-weeklyā€ to mean twice a week - which is not what we mean when it comes to rapamycin).

Iā€™d still add daily taurine a few days after the Rapa dose until the next Rapa dose. I read a recent study that taurine inhibits Rapamycin induced autophagy. Thatā€™s not what we want to do. I think it should be safe to start taking again after about 3 days (72 hrs) when it is past Rapaā€™s half-life (63 hrs).

In other words, does taurine reduce the effects of rapamycin? Do I understand that right?

RapAdmin, what is your current dose of Rapamycin?

I took 22mg of rapamycin (with GFJ~77mg enteric) at 12.30pm today. I felt a litttle tired about an hour later, but I think that is moreso because from 6.30am to 9.30am I was driving between Birmingham and Devon.

I am slightly drunk ATM, but that is not AFAIK rapamycin related, more related to beer and gin.

I am not wearing a CGM. I will do a blood test this week, but only one.

What detailed blood testing are you going to do? I just did 2.5 hours after dose; 72 hours after dose; and finally 2 hours before the next dose. Still waiting for results.
