I also have Rapacan but I’ve not started. I’m going to compare the potency the same way. Hopefully it is better.

I tested the SiroBoon one more time with 12 mg and grapefruit, this time in enteric capsules. After 17 hours I was at 1.9 ng/mL.

I got the Rapacan in the mail and tested 8 mg with grapefruit. My 2 hour mark was 30.1 ng/mL. I think I’m just going to throw the SiroBoon away.

Yes - I wouldn’t recommend people waste their time and money on SiroBoon. They are a 3rd or 4th tier Pharma company in India - so they are unlikely to be a company you want to rely on for any sort of healthcare product.

As a general rule of thumb, focus only on the top tier Indian Pharma companies that also sell generic medications in the US and Europe (i.e. have FDA approval for some significant number of their products). These are likely the best bets for getting a safe and effective product. I will also note that pricing in the US has come down so much now for sirolimus / rapamycin (with suppliers like CostPlusDrugs.com, and using the cost-reduction GoodRX.com service) that the pricing benefits or ordering from India is pretty minimal, and with shipping, and money transfer / wire fees you may not save any money at all.

Generally (and I encourage you do do your own research), the list seems to include these companies:

India is still the best place to buy most generic drugs that are expensive in the USA such as Bempedoic Acid and SGLT2Is. I even get my Telmisartan there as it’s still a savings.

I took 8mg of Rapacan last week and my 84 hour blood test came back at 13.0 ng/mL. That is like 4x higher than my Siroboon tablets… Now I feel like I have to drop my dose down by at least half! I was going to take it this week but I think I will wait till next week and start a new cycle with 4mg.

Before seeing this thread, I purchased a couple strips of Siroboon, so really appreciate you documenting your testing with this version of Sirolimus. Do you still have the packaging for your Siroboon? If yes, does it say each tablet is film coated?

The ones I received were manufactured April 2024 and the box says they are coated (see attached). I’m wondering if Kachhela improved their production by enterically coating now.

I don’t know for sure, but I suspect the large variance in the blood levels seen with Siroboon / Kachhela products is more a result of the poor quality control and not whether there is a film coating or not. But - do the blood test yourself and report back if you can. There are some pretty cheap testing options now: GetHealthspan - new Sirolimus level testing

They are not coated… you can take more but the results are wild unless you have a meal 3 hours before you take them.

Do you have one of your siroboon packages left to see if it says “film coated”? I’m curious if this is a recent addition

Read the thread. Don’t buy Siroboon when there are other better alternatives that are only marginally more expensive.

I did, and I will going forward. I’m trying to figure out the best use of the strips I have. And if there has been a change in quality, or at least the addition of enteric coating, I would think the community would be interested to know that.

It says coated. Only God knows what kind of coating it has.

I plan to crush all the siroboon tablets to use topically.

My gout was getting triggered 2 to 3 days after taking siroboon but not the rapacan brand. It makes sense because I’ve to take a lot of siroboon to get the same blood levels.

Wow what good effects did it have with all those negative side effects?

The point was I was taking too much at weekly doses, not that sirolimus is inherently bad.

Now at biweekly doses with occasional breaks and things are going much better.

I’m having a hernia operation tomorrow 1/9/25. I will skip my weekly dose of 7mg. based on your 7 week break from dosing.

I’m struggling to understand what the range should be in your blood for an anti-aging effect. My results below are 3 hours after taking my weekly 6mg of Dr Reddy’s from a US pharmacy (I have a prescription). It seems like from what I’ve been reading on here, my dose is too high? I’d love to have the collective’s thoughts.

I don’t find a 3 hr level useful. For weekly dosing, I’ve gone to a 50 hr level, and for every 2 week dosing a 100 hr level. The goal is around 3 ng/mL. In this fashion, if you are close to that, then you have 30% of the time with a level above 3 ng/mL, and 70% in recovery.
Doing a level in this fashion controls for absorption and metabolism, and I’ve not seen any excess side effects in my patients with taking this 30:70 approach, at least as stated above.

When Matt Kaeberline does 8 mg dose and tests 3 hours later he reported a 22 ng/mL .

I use that as a normal guide about 3.0 ng/mL per 1 mg rapamycin pill. I am finding that is my response with pure rapamycin (no GFJ).
