I tried lemborexant and find it amazing! In my N=1 experiement, micro-dosing 1mg lemborexant increased my deep sleep according to OURA ring.
However I read both lemborexant and rapamycin are motabolized by CYP3A4 enzymes. I’m concerned about potential drug-drug interactions. Is it safe to take rapamycin in the morning and lemborexant at night?

I take both of them and I haven’t find any side effects.
The extent of CYP3A4 inhibition varies depending on different drugs and the dosage.
Even after all the substance is metabolized, CYP3A4 needs time to “refill”, and the time varies from person to person.
I‘ve suffered from severe liver injury by longevity supplements last year, and the reason might be my CYP3A4 enzymes almost fully inhibited and the glutathione in the liver “depleted”.
The safest way is to test your AST/ALT/GGT regularly. Maybe at least once every 3 months, since liver usually needs 3 months to recover after injured.
