I am a fan of Physionic. He tells it like it is.
Urolithin A has effects that are non-existent or small.
No need to chase another supplement whose supposed benefits are questionable and is expensive if taken in the supposed therapeutic doses.

Seems to me that UA might be good at driving mitophagy and that doing cycles of a few weeks say 2-3 times are year could make sense.
I donât think weâd see better excise enhancing performance metrics from other mitophagy/autophagy compounds like rapa or say 3-5 day water fasts - even if I still believe they are helping a persons longevity phenotype.
So not sure that should be the bar for UA (even if I understand that the companyâs marketing and sales departments want to be able to use that type of data for the larger exercise and athlete market than the smaller longevity market).
If one is only doing cycles of a few weeks say 2-3 times are year the cost would be massively lower.

[Best Robert de Niro]: You talkinâ to me?
I hadnât watched the video when I posted, but Iâve watched it now. First, heâs talking about a different study: this 2019 one-month study of UA vs. mitochondrial gene expression. I was talking about the study referenced by cl-user: this 2022 four-month study on muscle function and metabolome. So one reason we came to different-sounding conclusions is that we were discussing different studies. (And I agree with Physionic that the 2019 study is a yawner).
Even granted that, our evaluations really arenât that far off. He says (23:28): âThat doesnât mean that UA is ineffective â just that it probably should be (based on this limited data) further down on the list of priorities. Once more data releases, Iâll update that stance.â We have more data, so he would have an updated stance. And even regarding the 2022 study, Iâm not exactly electrified, as I thought I had made clear:

Possible monkey wrench ⌠maybe we donât want our mitochondria to be too energetic?
[There are two types of people in this world: those who are lost and confused and know it, and those who are lost and confused and donât know it. ;-)]

With a bit more ATP perhaps the neurons will function better and people wont be confused.
There are lots of diseases caused by inefficient mitochondria. The situation is however complex. I am reading a lot about ot ATM.
This was a fruit fly study; we already have lots of data in rodents, which have the advantage of being mammals.

Itâs now been about ten days that my partner and I have been taking 500mg a day. Weâve both been having headaches, her much worse, but no lack of energy to speak of.
Itâs hard to tell if the headaches are connected to this. She gets them before her period, but itâs been earlier than usual for headaches. My headaches have been especially mild, but I never get headaches. So itâs notable in both cases.
As of now we both plan to push through. Itâs possible Iâve had a tiny bit more energy than usual. My partner says she hasnât noticed any change in energy, âless or more.â
Oh funny, she never has much gas but itâs been eye watering in the last three days. Itâs been a joke between us. Hard to tell what itâs related to. She said sheâs going to change some things up tomorrow.

The theory of mitophagy is that it is a process a bit like a spring clean where the cell removes some of the mitochondria that are not functioning well (AIUI by looking for ones with bad membrane potential) and then grows some more.
Hence
a) It is not something that must be done every day
b) There is an argument potentially for having stronger days for mitophagy perhaps by taking more than one dose.
I experimented two weeks ago with Aeternum and about an hour ago I took Mitopure (one sachet). Last tuesday I took 6mg of Rapamycin with a Pomelo (which means I probably have some Rapamycin effects at the moment).

In the last two days our headaches have subsided significantly. Mine is gone completely and hers mostly. Seems like it may have been due to sinus pressure.
I will say that Iâve been getting the slightest lip ticks, like a momentary involuntary tension in a small part of my lip. I recall getting this at some point early on with rapa but not for ages. Perhaps itâs related to the mitophagy.
In any case we continue to choose the red pill every morning (we enjoy the matrix reference) and will continue to report back.

Also AKG, Taurine, Gemfobrozil
- When it comes to AKG, NIA ITP tested this 4 years ago - still no results but probably doesnât work on their mice or effect is extremely small
- Taurine - works on mice but doesnât work on fruit flies - so may slow aging only in some animals
- Gemfobrozil, Urolithin A - not tested by NIA ITP yet - we also donât know what are interactions (if it works well with Rapamycin or not)

Why do you conclude this? The ITP website says testing is still in progress,, so the only thing we can conclude is that it didnât kill all the mice immediately.
Yadav didnât test it in flies. It worked in C. elegans and mice and showed good translational evidence in monkeys and thereâs short0-term human trials and epidemiology to support it. It didnât work in yeast, which the authors reasonably attribute to the fact that yeast donât have the same taurine synthesis machinery that multicellular organisms do.

Theyâve just answered, more than a month later:
Thank you for your inquiry! At present, our clinical studies have primarily focused on daily dosing of Mitopure Urolithin A. The dosing schedule, including days off or intermittent dosing, hasnât been a primary focus in our current research. As our understanding evolves and we conduct additional studies, weâll be sure to share any relevant findings.
We only offer our Mitopure challage test where you can see if you are a producer of UA and the differance in UA levels after a dose of Mitopure. You can check out our Mitopure Challange here: I have pasted a link below where you can learn more about our at-home test kits and apply for one.
https://challenge.timelinenutrition.com/ âââââââ
(typos not mineâŚ)
The website says: âWe have a limited amount of kits available. Apply now to participate! Currently available to US Residents onlyâ.

HiďźWhich do you think has greater health and longevity benefits, FMD fasting compared to Urolithin A

@sml491010
Short thoughts are:
-
FMD (and cheaper (and for me actually easier, water only fasting) is more broad based in its effects - both mitophaghy and the broader autophagy as well as whole range of other things like modulating mMTOR, IGF-1, training your body to be able to use ketone bodies, etc etc
-
UA is more targeted on mitophaghy. Itâs easier to take a pill. More expensive. But effects wonât be as broad based.
(Research risks with both too, though they seems quite ok in generally healthy individuals on both ends in short to medium term, long term less info)

Iâm a fan of four-day water fasts. But they will reduce lean muscle as well as fat so you need to balance it out with muscle building (my assumption). I donât know if UA has that same negative effect.

I have bought pomegranate juice which I drink, itâs not that bad. 1 L is made of ~2 kg pomegranates from concentrate, couldnât find fresh. PJ is in Bryan Johnsonâs nutty pudding. Might remodel gut bacteria over time to produce more Urolithin A, if it turns out bad I can just stop it.

The only bad that may come from it is the sugar content that they might add to it. If natural with no added sugar pomegranate juice is a healthy type of juice.

Yeah I made sure it had no sugar added, I skipped over those that did, I think drinking it with a straw like any fruit juice is probably a good idea as well.

Why would drinking with a straw be any different than drinking it directly from a glass? Donât both methods take it to the same place. Never heard of thisâŚLOL
