I’m very interested in this approach. How long do you leave between after the 4 daily doses?

My hypothesis is … The fasting mimicking diet research suggests that autophagy really kicks in after 4 days. So maybe a multiday pulsed dosing followed by a long gap could boost autophagy.Not sure my logic is sound but interested to hear everyone’s thoughts

Weekly dosing, with dog aging also moved to weekly, but they say it’s all guesswork

I break for 14 days usually after 4 mg doze during 4 days.

I am a 49 year old woman who is 5’2 and weighs around 130lb… I started testosterone therapy a few years ago (have not needed estrogen yet) and have been doing peptide stacks for the past 3 years (yearly) with CJC1295/Ipamorelin and Epitalon/Thymosin… I have had noticeable positive effects from peptides…I have also done BPC 157 and TB500 for injuries(which worked quite amazingly I might add)… I am an expedition hiker and still spend 100+ miles on the mountain/trails each year, I also do a combination of cardio and weight training 3-5 days a week…my only health condition is Hasimoto’s which I have had since I was 23 years old and has been managed with desiccated thyroid since my diagnosis…I am a registered nurse as my full time job and only participate in standard “western medicine” at work… I see a regenerative medicine physician as my primary care and really have no health problems… I started rapamycin today at 3mg and plan to do 3mg a week for 4 weeks then increase to 6mg per week… I have a few questions if anyone has advice or answers…

  1. Has anyone had regular thyroid testing, specifically TPO and if so did you notice a reduction in TPO? (My TPO has rarely ever been under 100 in my entire adult life despite medication changes and an absence of other thyroid pathology… I had a slight reduction with testosterone therapy, but I don’t ever “feel bad”, so I’ve just accepted my TPO will always be high, my CRP and RA factor have always been negative)
  2. Has there been any indication that sex and/or weight factor into dosing?
  3. Is there a consensus on the duration of rapamycin therapy before “taking a break” and if so how long should that break be? I’ve read many differences in opinion on this and am not sure if emerging research has any new information that I may not have come across
  4. I’ve genetically had white hair growing in starting at 16 … my mother is Scandinavian and it’s a family trait… has anyone had repigmentation of their hair? I doubt mine will change since this is definitely a genetic predisposition, but I’m curious
    Thank you for any insight… I will have my next labs drawn in a few months and will post an update if my TPO goes down… just looking through the posts, I think there is a majority of men on the thread, so I will update on changes I see from a woman’s perspective… I am peri-menopausal still having normal menstrual cycles-I’m adding this for perspective since post-menopause adds significant changes to most women’s bodies… thanks again!

G’day Jason, I presume you are going 4.6.4.6.4.6… progressively, keeping you just under 7mg given the 2.6 day half-life of Rapa. Are you taking a break regularly? I’m glad you dropped the dosage down. You can certainly get too much of a good thing.

I’m keeping under 5mg for a three weeks and then resting for a three weeks. I feel like this is giving me enough of the important autophagy benefits without any annoying ulcer side-effects.

To bad we didn’t connect when I was down under in Sydney end of October. I had a blast with the locals.

So I started off at 6 mg… in August 2020. Great results… went high after a year and a half… bad results. Went low to 4 mg… not so good results either… benefits slowed skin was first sign of problems… dried out.

I have returned to my original dose… and adopted a straight 6 mg weekly. Seems to be working.

No I have never taken a break… will be 5 years in August 2025.

I would be a walking ulcer if I’d crashed thru 5 years uninterrupted! That’s an impressive commitment. Power to you!

Yeah, I’ll be traveling a lot more too in a decade, so maybe we can all meet up Down Under.

The great thing about Rapamycin is that it (probably) gives us an extra healthy decade where we can truly live life unfettered!
