Yes, Pomelo is even more potent than grapefruit at inhibiting CYP3A4. I can’t seem to find them in the stores that often though and like to stick to a routine.

I live on the edge of inner city birmingham with a wide range of shops serving different ethnicities and ordinarily stocking pomelo and other otherwise unusual fruits.

I see them frequently in the Chinese markets (Chinatown, other areas) in San Francisco. But I’m fine with grapefruit, so don’t usually buy them.

If grapefruit has such a big effect in absorption of RAPA (as an example) wouldn’t it interfere with other medicines and even various nutrients also?
Since i learned that grapefruit multiplies the absorption of RAPA you can rest assured, I’ll never touch it again, other than when I take RAPA. You don’t need to be a doctor to understand that it messes up your system.

I was looking for something more specific. i.e lets say i eat grapefruit as desert will it affect the way nutrients from the meal are absorbed? and is there any case that it is a good idea to eat grapefruit for certain purpose (other than to increase medicine absorption). I’m more interested in food and nutrient digestion effects if any.

Just got my results. The lab toxicologist panicked and wanted to admit me into the hospital. My 26mg with 3 Grapefruit after 2 hours yielded 27.2ng/mL and after 24 hours 9.7ng/mL and I’ll do one more test after 7 days.

Much that I will stick to my 6mg (or 5 depending on packet) plus Pomelo, there has been an N=1 experiment with more than you took and we have not seen any immediate problems although I would expect mTOR to be inhibited for quite a while.

Based on the Blueprint Protocol, I think 27.2 is in the correct target range. Maybe I will drop it back a notch to 24mg / 6mg with at least 500ml of GFJ or 2 grapefruit or 1 pamelo plus 1x 100mg aspirin. I still do not like GFJ or grapefruit that much. Pamelo is far more palatable. Now I will see how my arthritis goes after 6 months.

After 1 week my blood level is 2.4ng/mL. The next day I took 6mg with 2 grapefruit and after 75 minutes it yield 3ng/mL.
I’m waiting for the enteric capsules to arrive and I’ll have these pills inside the capsules and see what happens.

Be careful about your own CYP enzymatic make-up!
About 12 years ago I was on simvastatin and developed community pneumonia. I went to a local clinic. He put me on erythromycin, and never checked what else I was on. Well, simvastatin + erythromycin are strong inhibitors of the CYP3A4. One was fine—two was definitely not.
The next afternoon at work I started feeling horrid and did some research on erythromycin and saw the two really were incompatible. I had a multistrip in my clinic (I’m an RN) and the indicator for blood in urine when solid black, instantly. But there was no blood in my urine. There was however, myoglobin. I had Rhabdomyolysis! Luckily there appears to be no major damage done. Had I kept taking the erythromycin, in a day or three, my liver would have been destroyed.
The pharmacist at CVS said: “The computer should have pinged me when it saw that you were on simvastatin.”
I later found that exactly the same thing happened to my niece. Therefore, I think it’d be wise that I consider going very slowly with the grapefruit juice. Anyone else with similar experience?

The last result is a bit unusual. I had 500ml of GFJ from the bottle. About 1.5hr later I had the 100mg aspirin and some breakfast with my wife consisting of half a garlic bread with avocado. After that on the 2hr mark I had 13x 2mg pills doused in hemp oil. 24.5 hours later I returned a result of 18.2 ng/L. I didn’t even feel tired.
Previously 2 weeks ago, my 24 hour test yield 9.7ng/L. And I felt fatigue on the first day. The only different this time is I had more food, and GFJ from the bottle and took the rapamycin around 10.30am instead of 8am.
Next week I’ll take 12mg encased in enteric capsules blanks I bought and try to repeat what I did this week.

See how I go this week with my blood test. The only question is should I put the hemp oil into the capsules or take them separately? I’m planning to have it separately to maintain the integrity of the capsules.

It seems like a lot of work to replicate the results we would normally get by using Rapammune, Zydus or Biocon brands.
To me, Siroboon isn’t worth the effort.

I think the problem is Australia Border Security not allowing me to order anymore rapamycin until August. Anyway it is interesting and keeps my mind busy lol.
